神经黑客(Neurohacking),有时也称为大脑黑客(Brain hacking),是健康个体优化大脑功能和提高认知能力的尝试。

现代神经黑客运动自20世纪80年代以来就存在。过去数百年中,草药补充剂一直被用于增强大脑功能3。在该领域缺乏研究的短暂时期之后,自本世纪之交以来,有关科学出版物的普及提高了大众对神经科学的认识4。因此,公众意识到了大脑的可塑性及其改善的潜力,[6]神经黑客开始重新引起人们的极大兴趣5。自2011年起,在家庭和通过自己动手(DIY)的神经黑客活动再次流行起来2。
简单的神经黑客包括利用促智补充剂(Nootropics)来增强大脑功能6。“Nootropics”一词最早由布加勒斯特大学的罗马尼亚化学家Corneliu Giurgea于1972年提出7。此外,据美国FDA,可以植入更复杂的医疗设备来治疗心理障碍和疾病8。如今,通过处方和柜台都可以买到各种益智药或补充剂9。

1.促智剂
促智剂(Nootropics)是任何可增强大脑功能的化合物10。尽管许多是由身体自然产生的,但通常需要可摄入的补充剂来提高血液中这些化合物的浓度,以产生显著的效果。促智剂可以进一步分为两类:合成Nootropics和天然Nootropics5。
1.1.合成促智剂:指实验室生产的益智药剂,包括吡拉西坦(一种常见的脑血管病药)11。合成促智剂用于三个不同的连接点5:

2.程序性训练练习
程序训练方法加强了神经元之间的联系。例如,大脑训练游戏自2000年代以来就一直存在。PositiScience、Lumosity和CogniFit等公司开发了旨在改善用户大脑功能的视频游戏2。这些大脑训练游戏通过在理解技能中添加类似游戏的特征来提高神经能力12。
3.电流传输
电流通过大脑传输有三种方法:脑深部刺激(DBS)、经颅磁刺激(TMS)和经颅直流电刺激(tDCS)13。
3.1.深部脑刺激(DBS):DBS涉及将一种电气设备或神经刺激器植入大脑14。 神经刺激器是一根尖端有电极的细线,低水平的电流通过大脑传导。植入电极的位置取决于所治疗的神经系统疾病。Neuralink公司的DBS设备将包括“多达3072个电极,分布在96条线上”,并且植入丝线的程序将与LASIK眼科手术一样无创。
3.2.经颅磁刺激(TMS):TMS通过一个小的电磁线圈向左额叶皮层发送短脉冲的磁能。一些研究发现,TMS可以改善认知和运动表现15。其他研究调查了经颅磁刺激与其恢复丢失记忆能力之间的关系。
3.3.经颅直流电刺激(tDCS):脑细胞或神经元通过神经元之间的间隙或突触发出化学信号。当学习一项新技能或主题时,参与理解特定主题的神经元会更容易地发出信号。需要较少的电流来向神经元发出信号,以分泌通过突触间传输的化学物质。tDCS包括通过放置在头部上的阳极和阴极运行非常低的电流(小于2mA)。研究表明,阳极周围的脑功能得到改善,阴极周围的大脑没有变化或功能下降16。
本文侧重于提高大脑功能体验的“营养神经黑客”(Nutritional Neurohacking),通过使用促智补充剂,有助于优化大脑健康和功能。
1.心理健康:

增强人类体验是神经黑客的另一个应用。方法包括简单的大脑训练游戏、促智剂和脑电刺激。例如,咖啡因是全球最受欢迎的促智药,可提高记忆力、社交能力和警觉性(详见下文)。
3.信息检索:
神经黑客的第三个主要应用是从大脑中检索信息。这通常涉及使用脑机接口(BMI),一种测量大脑电信号的设备17。

个体的神经黑客目的可能部分取决于年龄。例如,老年人通常关心神经保护和保持健康的大脑功能,而年轻人通常更专注于通过促智剂和其他大脑黑客技术来提高心理表现和耐力。
神经黑客的综合方法依赖于各种互补的策略,包括饮食、生活方式调整,促智药或补充剂的使用,大脑训练活动和游戏,以及旨在提高大脑健康的神经技术(如电刺激装置)2。
认知测试通常用于评估几个特定的认知领域,包括如下20:
本文中的营养素和植物药(提取物)已被证明可以改善健康人大脑或认知功能的一个或多个方面。需要注意的是,由于本文专注于对健康受试者认知功能的研究,一些通常与大脑健康相关的营养素(如甲钴胺、低剂量锂等)可能不会出现在这个列表中。这是因为支持这些营养素的研究通常涉及有潜在神经精神疾病的人,而不是健康人。
值得注意的是,“营养神经黑客”应被视为一种整体方法,强调健康饮食和整体营养,并结合有针对性的营养补充。例如,2022年11月发表的一项随机安慰剂对照试验发现,在习惯性健康饮食的参与者中,补充12周的维生素B族成员、假马齿苋和银杏叶可以改善注意力表现和情绪的某些方面,但在不健康饮食的参与者中则难于产生预期的效果22。

1.获得最佳大脑表现:

1.1.假马齿苋:
假马齿苋是印度阿育吠陀医学传统药草,可以增强记忆力、减少焦虑和调节血糖水平23。在健康成年参与者中进行的临床试验表明,与安慰剂相比,每天300mg假马齿苋提取物持续90天至12周,可以提高记忆力和情绪测试、视觉处理速度、学习率和一般认知功能的得分24-26。
在一项包括60名医学生的随机安慰剂对照试验中,连续六周每天两次服用150mg假马齿苋标准提取物,可改善注意力和工作记忆测试的表现27。一项包括了60名老年参与者的试验发现,与服用安慰剂的人相比,每天300或600mg的剂量持续12周,在记忆、注意力和认知处理测试中表现更好,表明工作记忆有所改善28。假马齿苋还抑制乙酰胆碱酯酶活性,这种作用会调节神经递质乙酰胆碱的可用性,并与神经保护有关29。此外,在停止补充后四周的随访测试中,假马齿苋对认知的益处持续存在28。一项对28名55岁以上的健康成年人进行的随机安慰剂对照试验发现,与给予安慰剂加认知训练的人相比,那些每天服用320mg假马齿苋提取物、持续12周,每周进行3小时认知训练的人,反应速度较慢,但认知测试的准确性更高30。
一些证据表明,假马齿苋可能具有即时、短期的促智作用,这使得它针对那些对大脑黑客感兴趣的人很有吸引力。在一项有17名健康参与者的小型交叉试验中,与安慰剂相比,在单剂量320mg或640mg标准化高浓度假马齿苋提取物后一小时和两小时进行的多任务认知测试中,表现有所改善。在这项试验中,假马齿苋还降低了唾液皮质醇(体内主要应激激素)的水平,并对情绪产生积极影响,这表明假马齿苋可能在一定程度上起到适应原(增加应激适应性的物质)的作用31。在一项类似的试验中,24名健康志愿者在服用一剂320mg假马齿苋提取物后不久,在重复认知测试中的持续表现优于640mg假马齿苋或安慰剂32。
1.2.蓝莓:
蓝莓以其富含花青素的多酚类物质而闻名。大量研究和临床试验表明,蓝莓提取物和花青素可以增强健康人的认知功能。在一项系统综述中,包括10项在健康成人和儿童中使用蓝莓提取物的对照试验和5项在轻度认知障碍的老年人中使用的对照试验,发现主要的认知效果是记忆改善33。另一项对11项在无认知障碍或轻度认知障碍老年人中进行的试验的综述发现,蓝莓干预可以改善记忆、工作记忆,执行功能、处理速度、注意力以及脑血流量34。一项对六项随机对照试验的荟萃分析发现,蓝莓补充剂显著改善了一项工作记忆测试的表现35。
一些临床证据表明,蓝莓补充剂对认知功能有敏锐的益处。一项针对35名健康中年人的安慰剂对照交叉试验发现,单次服用25g冻干野生蓝莓粉可以改善认知功能测试的表现,尤其是那些涉及更高要求的心理任务的测试36。在一项包括30名健康大学生的安慰剂对照试验中,那些从蓝莓和葡萄中摄入600mg多酚的人在补充后90分钟开始的长时间、高要求的心理测试中表现更好37。
1.3.类胡萝卜素:
叶黄素和玉米黄质是类胡萝卜素家族中的脂溶性营养素。它们高度集中在黄斑,黄斑是眼睛视网膜中负责中央视觉的区域,在那里它们可以保护自身免受氧化应激和短波蓝光的损害38。它们在脑组织中的积累也比其他类胡萝卜素多,因此,叶黄素和玉米黄质占大脑类胡萝卜素含量的66–77%,在减少自由基和减轻神经炎症方面发挥着重要作用39。多项研究表明,在健康的年轻人和老年人中,这些类胡萝卜素的摄入量和浓度越高,患阿尔茨海默病的风险越低,认知功能越好38,40。健康人的脑成像对老年人研究发现,在一年内每天服用10mg叶黄素和2mg玉米黄质,可以增加学习和休息状态(一种神经可塑性的测量方法)下的大脑活动,以及对补充剂反应更好的个体的大脑结构、大脑灰质(主要由神经细胞体组成)体积下降较少41-45。
一项对9项随机对照试验的荟萃分析发现,在健康成年人中,补充叶黄素、叶黄素加玉米黄质、虾青素或β-胡萝卜素可以改善各种认知指标46。例如,一项在59名18-25岁的年轻健康成年人中进行的随机安慰剂对照试验比较了13mg或27mg叶黄素、玉米黄质,以及一种名为消旋玉米黄质(Meso-zeaxanthin)的相关类胡萝卜素和安慰剂。研究人员发现,六个月后,补充类胡萝卜素会降低炎症细胞因子(白细胞介素-1β)的水平,增加脑源性神经营养因子(BDNF)的水平和抗氧化能力,并在记忆、注意力和处理速度测试中表现更好47。类似地,一年后对51名18-30岁的健康受试者进行的安慰剂对照试验,发现可改善记忆力、推理能力和复杂注意力的测试48。在一项随机安慰剂对照试验中,90名40-75岁的健康参与者报告了记忆力、注意力或专注力方面的问题,10mg叶黄素加2mg玉米黄质,每天一次,持续六个月,改善了与学习和记忆相关的某些认知测试的表现39。一项针对91名黄斑类胡萝卜素浓度较低的18岁至60岁健康人的试验发现,一年内补充10mg叶黄素、2mg玉米黄质和10mg消旋玉米黄质比安慰剂更能提高记忆测试的表现49。
1.4.姜黄素:
姜黄素具有抗炎和减少氧化应激的特性,在体内具有广泛的药理作用,并能穿过血脑屏障,使其具有保护神经系统的潜力50。一项2021年的荟萃分析使用了8项随机对照试验的数据,共有389名参与者,其中4项试验针对健康老年参与者,二项试验针对阿尔茨海默病患者,二项研究针对精神分裂症患者。纳入的试验使用了不同剂量的不同类型的姜黄素制剂。分析发现,总的来说,姜黄素治疗至少八周可以改善工作记忆51。另一项荟萃分析表明,姜黄素在改善健康老年人的认知功能方面可能比减轻阿尔茨海默病或精神分裂症患者的症状更有用52。
一项随机安慰剂对照试验在60名健康老年志愿者中进行,他们接受了400mg提供80mg姜黄素的脂质制剂,一小时后发现该制剂改善了注意力和记忆力。此外,连续四周每天服用姜黄素补充剂可以改善工作记忆和情绪测试的表现53。在一项安慰剂对照试验中,80名50至80岁的参与者每天服用相同剂量和形式的姜黄素,持续12周,改善了工作记忆,降低了疲劳54。然而,另一项安慰剂对照试验在39名年龄在45岁至74岁之间的健康男性和女性中进行,发现12周后服用更高剂量(每天2000mg)的相同姜黄素制剂(提供400mg姜黄素)对认知没有益处55。
在40名年龄为51-84岁的受试者中,与安慰剂相比,每天两次服用90mg不同生物可利用形式的姜黄素,持续18个月,可以提高记忆力和注意力测试的表现,还可以减少大脑中异常淀粉样蛋白和tau蛋白(与阿尔茨海默病有关的蛋白质)的积累56。然而,一项包括96名健康老年参与者的试验发现,在12个月内保持认知功能方面,每天三次500mg的另一种姜黄素制剂并不比安慰剂好57。
1.5.野燕麦:
绿燕麦历来被用于治疗焦虑和失眠,临床前证据表明它可以对大脑功能产生积极影响58。在实验室研究中,野燕麦提取物被发现能抑制两种对心理和认知健康有作用的酶:单胺氧化酶-B(MAO-B)和磷酸二酯酶(PDE)59。抑制MAO-B会导致多巴胺等神经递质水平升高,而PDE抑制会导致神经递质信号传递的变化60,61。
在一项针对20名健康参与者的对照试验中,与安慰剂相比,单次800mg剂量的绿燕麦提取物改善了工作记忆、处理速度和注意力测试的表现,并改变了大脑电信号62。一项针对自我报告感觉记忆力下降的健康中年人的试验也显示,在服用800mg(而不是1600mg)绿燕麦提取物后的6小时内,他们在速度、记忆力和执行功能的测试中,认知能力得到了显著改善63。另一项试验包括132名健康个体,他们每天服用430mg、860mg或1290mg绿燕麦提取物或安慰剂,持续29天。补充第一天的认知测试显示,1290mg绿燕麦对工作记忆和多任务处理有急性益处,在第29天,服用430 mg或1290 mg的人有了更大的改善58。然而,在37名参与安慰剂对照交叉试验的健康老年人中,每天1500mg绿燕麦提取物,持续12周,未发现对认知功能测试得分有影响64。
1.6.猴头菇:
猴头菇(又称狮鬃菇)是一种药食同源的菌菇。据报道,它对健康有广泛的益处,包括增强大脑功能,缓解焦虑、抑郁和疲劳65。在一项针对34名50岁以上健康人的随机安慰剂对照试验中,连续12周每天服用4粒800mg的猴头菇粉胶囊,可提高MMSE评分,MMSE是一种衡量认知状态的指标66。其他临床研究表明,猴头菇可能有利于改善轻度认知障碍患者的认知功能67,并有利于保持早期阿尔茨海默病患者的认知能力68。
1.7.中链脂肪酸:
虽然最常见的脂肪由长链脂肪酸组成,但中链甘油三酯(MCTs)由中链脂肪酸构成,如从椰子油或棕榈油中提取的辛酸和癸酸。MCTs可以直接代谢为能量或转化为酮体,为脑细胞提供能量。研究发现,食用强化MCTs的食物可以改善虚弱和认知受损的老年人的认知功能69,70。一项为期四周的对照试验包括30名健康的年轻人,发现与服用安慰剂的人相比,服用每天提供12或18g MCTs饮料的人在2-3周后评估执行功能和注意力的测试中表现更好71。在一项对照试验中,32名健康的老年受试者在饮食中添加MCT,从每天6g开始,经过三周的时间增加到每天18g,而31名类似的受试者则不服用MCT。三个月后,补充MCT的参与者改善了行走平衡,可能表明脑-肌肉交流得到增强72。
MCTs也可能对大脑功能产生直接的积极影响。一项对照交叉试验发现,20名健康的老年志愿者在吃了一顿富含MCT的饭(提供19.9g MCT)后,在注意力的认知测试中表现比吃了富含长链甘油三酯的饭更好73。一项针对19名60岁以上认知健康个体的类似试验发现,与不含MCTs的膳食相比,含20g MCTs膳食后的注意力、执行功能和整体认知功能得分更高74。
1.8. 欧米伽3脂肪酸:
ω-3脂肪酸(EPA和DHA)集中在大脑中,有助于维持神经细胞膜的流动性和功能,减少神经炎症和大脑氧化应激75。 DHA是大脑中最丰富的脂肪酸,与神经生长、功能和可塑性有关,这使得它在营养大脑的研究中显得特别有趣76;除此之外,DHA似乎有助于B族维生素(维生素B12和叶酸)对整体认知的重要作用,并促进脑磷脂(丝氨酸磷脂)的合成77,78。观察性研究表明,大量摄入鱼类、EPA,尤其是DHA的摄入可以减缓大脑衰老,支持健康的认知功能79,80。最近的证据表明,与磷脂酰胆碱结合的DHA比其他形式的DHA更有效地融入大脑,具有更强的抗炎和抗氧化作用75,81。
一项临床试验评估了ω-3脂肪酸水平与认知功能之间的关系,该试验包括来自Framingham心脏研究的2183名无痴呆症患者。更高的ω-3指数(定义为红细胞中DHA和EPA的浓度)与海马体积增加和更好的抽象推理有关。EPA和DHA分别维持了这些观察结果。在一项检查APOE-e4基因型非携带者的亚组分析中,表明对阿尔茨海默病易感性增加的遗传标志物、较高的DHA浓度或ω-3指数得分与较大的海马体积有关。相反,在APOE-e4携带者中,较高的EPA浓度与更好的抽象推理有关,较高的ω-3指数与较低的白质高信号负荷有关,这可能与某些脑血管问题的较低风险相对应82。
尽管临床试验的结果好坏参半,但一项对15项随机对照试验数据的荟萃分析发现,补充DHA,无论是否添加EPA,都能改善认知能力,尤其是对那些有轻微记忆问题的人。这些益处需要每天大于1g的EPA/DHA组合剂量和大于580mg的DHA剂量才能显著83。例如,在一项安慰剂对照试验中,176名18至45岁的健康成年人的DHA饮食摄入量较低,在六个月的时间里,每天补充1.16g DHA可以提高反应时间和记忆力的测试成绩84;然而,在一项针对18-35岁健康参与者的试验中,低剂量450mg DHA加90mg EPA或200mg DHA加300mg EPA,持续12周,并不能改善认知能力或情绪参数85。此外,荟萃分析后进行的两项随机安慰剂对照试验发现,尽管剂量和试验持续时间足够,但补充DHA和EPA对健康成年人或有自我报告记忆问题的老年人的认知功能没有影响86,87。
一项对照试验包括285名因冠状动脉疾病而有认知障碍高风险的受试者,研究了高剂量EPA和DHA的可能益处;在补充1860mg EPA和1500mg DHA 12个月后,发现语言流利性、语言、记忆和视觉运动协调性都有所改善,这些改善一直持续到试验结束时的30个月88。
有趣的是,并不是所有的临床试验都表明DHA比EPA更具有益智功效。在一项针对310名50岁以下健康成年人的随机安慰剂对照试验中,那些连续26周每天服用900mg EPA加360mg DHA补充剂的人在记忆速度和准确性方面的所有测试中都比服用900mg DHA加270mg EPA或安慰剂的人有更大的改善89。
1.9.吡咯喹啉醌(PQQ):
PQQ是一种关键的营养素和强效抗氧化剂,有助于多种细胞功能,如线粒体生物发生和基因表达90。临床前研究表明,PQQ具有神经保护特性,并已被发现支持神经元生长,减少活性氧(ROS)的产生,抑制谷氨酸诱导的中风91-93。临床证据表明,PQQ有助于改善健康人的认知功能。
在一项针对41名健康受试者的研究中,每天一次服用20mg PQQ或安慰剂,持续12周,服用PQQ的受试者在注意力和记忆的认知测试中得分显著提高,前额叶皮层的血流量也有所改善,与安慰剂相比94。一项对照试验将64名40-80岁的健康受试者随机分组,每天接受21.5mg PQQ或安慰剂治疗,发现服用PQQ的受试者记忆力、反应时间、注意力和执行功能方面都有显著改善95。另一项研究发现,与安慰剂相比,20-40岁的成年人每天服用20mg PQQ,持续12周,8周后处理速度和执行速度显著提高96。
1.10.丝氨酸磷脂:
可以由磷脂酰胆碱或磷脂酰乙醇胺合成,但更容易由富含DHA的磷脂酰胆碱和磷脂酰乙醇酰胺产生77,97。磷脂酰丝氨酸支持认知功能,如短期和长期记忆、学习、注意力、解决问题、语言和交流,以及神经肌肉活动,如快速运动反应和反射。大脑的磷脂酰丝氨酸含量似乎随着年龄的增长而减少97。在有自我报告记忆问题的老年人中进行的几项临床试验发现,每天补充300mg丝氨酸磷脂可以改善记忆和认知功能的其他方面98-101。此外,在18名健康男大学生中进行的一项安慰剂对照交叉试验发现,在一项名为“连续七次”的认知测试中,每天400mg丝氨酸磷脂持续14天,可以提高速度和准确性。在认知测试中,测试对象被要求从100开始,以7为一组向后数102。
1.11.芳香药草:
迷迭香(Rosmarinus officinalis)、留兰香/绿薄荷(Mentha spicata,Spearmint)和欧薄荷(Mentha piperita,通称薄荷)对大脑功能的影响已被研究。据认为,迷迭香酸(Rosmarinic acid),一种在这些和其他薄荷科植物中发现的芳香酚类化合物,可能通过其穿越血脑屏障并发挥抗炎和清除自由基作用的能力来促进神经保护103。此外,临床前研究表明,迷迭香酸和来自迷迭香、绿薄荷和薄荷的精油与神经递质受体相互作用,可能抑制乙酰胆碱酯酶的活性,乙酰胆碱酯酶是一种分解乙酰胆碱酯酶的酶104,105。
1.11.1.迷迭香:
在一项对80名健康成年人进行的对照试验中,与摄入等量的白开水相比,摄入250ml注入迷迭香的水可以改善认知能力,增加大脑的摄氧量106。一项对28名老年人进行的安慰剂对照交叉试验发现,在评估记忆速度的测试中,单次服用750mg迷迭香叶粉 (这一剂量被认为反映了可行的烹饪摄入量)后的6小时内,记忆速度有所改善,但在服用6000mg迷迭香叶粉后的数小时内,记忆速度恶化107。
还研究了迷迭香的持续使用对认知的影响。在一项对44名健康成年人进行的随机安慰剂对照试验中,发现在63岁以下的一组参与者中,服用含有迷迭香、鼠尾草和香蜂草的酒精提取物两周可以改善短期记忆,但对年龄较大的参与者则没有效果108。在78名健康大学生的随机安慰剂对照试验中,发现每天两次500毫克迷迭香,持续一个月,可以改善记忆力、情绪和睡眠109。然而,在一项试验中,40名低能量的健康年轻受试者在接受认知测试前一小时服用1700mg迷迭香与2000mg黑胡椒或安慰剂,未发现草药组合会影响警觉性或认知能力。有趣的是,参与者戴着鼻夹,以消除迷迭香可能产生的芳香效应110。
芳香疗法是实现迷迭香和其他芳香草药对大脑有益的另一种可能方法。一项针对144名健康志愿者的对照试验报告称,迷迭香精油可以扩散到大气中,改善记忆力和警觉性111,112。在一项包括20名健康志愿者在内的试验中,在散发迷迭香香气的环境中进行认知测试,可以提高速度和准确性113。
1.11.2.留兰香/绿薄荷:
一种富含迷迭香酸的留兰香提取物已成为研究其潜在益智作用的几项临床试验的主题。在一项针对142名年轻健康参与者的安慰剂对照试验中,那些在90天内每天服用900mg标准化为含有14.5%迷迭香酸和24%总酚类化合物的留兰香提取物的人,在测量注意力方面的认知测试中的得分比那些服用安慰剂的人有更大的改善114。在补充相同剂量的高迷迭香酸留兰香提取物后,反应敏捷性,即对刺激快速有效反应的能力也得到了增强,但在安慰剂后没有115。在一项安慰剂对照试验中,60名患有年龄相关记忆障碍的老年参与者,同样的治疗也被发现可以改善一些工作记忆测试以及自我报告的入睡能力116。
1.11.3.薄荷:
在对24名健康年轻参与者进行的安慰剂对照试验中,在单次100ml剂量的薄荷精油胶囊后的3小时内,他们在高要求认知任务中的表现得到改善,长时间认知活动后的精神疲劳有所减少104。在一项对144名健康参与者进行的对照试验中,发现使用薄荷精油进行芳香疗法可以提高记忆力和警觉性111。
2.专注和注意力,缓解脑雾:
可以缓解脑雾(思维不清晰、脑子不畅),增强注意力和任务执行。
2.1.镁:
镁参与许多细胞活动,包括能量生产和蛋白质和DNA的合成。通过参与并促进其他离子穿过细胞膜的运输,镁也有助于调节肌肉细胞收缩和神经细胞兴奋性,保护神经细胞免受可能导致神经细胞死亡的过度兴奋117-119。尽管血镁水平趋于正常,但衰老与体内总镁含量下降有关117。有研究认为,较低的镁浓度会导致慢性炎症和氧化应激,并且是许多与年龄相关的健康问题的一个因素117,120。

特别是L-苏糖酸镁已被证明比其他形式的镁更容易穿过血脑屏障,临床前研究表明,它具有神经保护作用,减少神经炎症,抑制脑组织中的β淀粉样蛋白沉积,增强大脑功能,减缓认知能力下降117,124-126。一项随机安慰剂对照试验,包括44名受试者,年龄在50至70岁之间,有记忆和注意力不集中、焦虑和睡眠问题的患者发现,12周的L-苏氨酸镁补充剂,根据体重每天1500至2000mg,相对于基线和安慰剂,显著改善了整体认知能力127。
2.2.鼠尾草:
鼠尾草(Salvia officinalis和其他品种)是一种富含多酚的植物,历史上一直被用于改善记忆、加快感官、增强大脑功能和延缓认知衰退128。鼠尾草的活性成分已证明具有减少炎症信号、降低氧化应激、调节神经递质水平、减少淀粉样蛋白诱导的神经毒性等作用,以及增加BDNF的释放128-130。在健康年轻人中进行的几项临床试验表明,不同剂量的鼠尾草提取物(S.officinalis或S.lavandulaefolia)可以对记忆和注意力以及情绪产生短期的积极影响128。
在一项针对26名健康运动员的安慰剂对照交叉试验中,在疲劳运动前两小时服用单一600mg剂量的鼠尾草提取物,其中含有S.officinalis和S. lavandulafolia的化合物,也发现可以降低感知耗力的运动水平,提高运动后的记忆测试分数131。一项针对65岁以上成年人的安慰剂对照交叉试验比较了单剂量(167mg、333mg、666mg或1332mg)标准化鼠尾草提取物在给药后1、2.5、4和6小时对认知能力的短期影响。试验发现,在年龄较大人群中,333mg的鼠尾草能最大限度地改善记忆力,并提高注意力测试的分数132。长期使用鼠尾草也可能对认知有好处。在一项对94名健康参与者进行的随机安慰剂对照试验中,与服用安慰剂的参与者相比,服用600mg含有相同二种鼠尾草成分的提取物的参与者在治疗后2小时和4小时进行的一些记忆测试中得分更高;经过29天的日常治疗,记忆力的改善更加明显133。
2.3.芒果叶(提取物):
研究了芒果叶及其活性成分芒果苷(Mangiferin)对脑功能和记忆的影响。芒果苷已被证明可以增强大脑中的电信号,并通过减少炎症和氧化应激、支持线粒体功能、调节神经递质活性和保护BDNF发挥神经保护作用134,135。动物研究结果表明,芒果苷可以保护大脑免受睡眠剥夺、衰老和毒素135。在一项随机、安慰剂对照、交叉试验中,包括70名年龄在18岁至45岁之间的健康参与者,在服用300mg标准芒果叶提取物30分钟、3小时和5小时后,整体认知功能有所改善。与安慰剂相比,当补充芒果叶提取物时,参与者表现出更好的记忆力、注意力和执行复杂任务的能力136。
2.4.积雪草:
积雪草(或称雷公根)是一种亚洲药用植物,通过其减少炎症、氧化应激和线粒体功能障碍的能力,已证明具有神经保护作用,同时增加神经元连接137,138。在一项针对20多岁成年人的试验中,发现一种标准化的积雪草提取物可以提高血液胆碱水平139。
在一项安慰剂对照试验中,包括28名健康的老年参与者,连续两个月每天750mg 积雪草提取物改善了工作记忆,并增加了认知任务期间的脑电活动,但每天250mg和500mg都没有这些效果140。一项包括五项随机对照试验的荟萃分析发现,积雪草可以提高警觉性,减少愤怒,从而改善心理功能;然而,现有的研究不足以得出关于其增强认知能力的结论141。
2.5. 二甲氨基乙醇(DMAE):
DMAE是一种胆碱前体,可优化大脑乙酰胆碱的产生,已证明其具有清除自由基的作用142。一项针对80名情绪障碍受试者的安慰剂对照试验发现,含有DMAE的多种营养补充剂可改善情绪,并在某种程度上改变了脑部电活动,表明注意力增加143。

在健康中青年志愿者的安慰剂对照交叉试验中,发现单次剂量为100、200和400mg的西洋参标准化提取物可在1、3和6小时后改善认知能力145,146。例如,在一项包括61名健康青年和中年参与者的安慰剂对照试验中,单次200mg剂量的标准化西洋参提取物在补充后4小时和6小时的工作记忆和注意力测试中表现增强;此外,连续两周每天服用同样的200mg补充剂可以减少精神疲劳,增强情绪,并进一步提高认知测试成绩;而且人参似乎可以调节肠道微生物组的组成,这可能有助于其认知效果147。
韩国参也一直是健康个体临床试验的主题。据报道,在健康的年轻人中,单次服用200-400mg的高丽人参可以改善认知功能,减轻高要求脑力任务时的精神疲劳148-150。一项针对51名健康参与者的试验发现,与安慰剂相比,连续八周每天服用1000mg高丽人参不仅能提高整体认知功能得分,而且还能增加灰质(主要由神经细胞体和其他细胞组成的脑组织)的体积151。在一项交叉试验中,包括50名具有高度职业压力的健康个体,在1天、5天和12天后,每天服用418mg人参皂苷富集制剂的高丽人参比每天服用768mg未富集的高丽人参粉或安慰剂更有效152。
3.缓解脑疲劳、增强认知耐力:
可以强化认知耐力,避免普遍的精神疲劳。
3.1.乙酰L肉碱:
L-肉碱是细胞脂肪代谢和线粒体能量生产所必需的氨基酸。它在体内制造,储存在骨骼肌、心肌和大脑等代谢需求高的组织中。乙酰L肉碱是一种穿过血脑屏障的肉碱,因此可以通过增强神经生长因子、神经递质和神经激素的产生来调节神经活动153。此外,乙酰L肉碱可以帮助恢复神经细胞膜的完整性和功能,增强线粒体功能,并防止神经毒素154。
一项随机安慰剂对照试验包括96名70岁以上且患有疲劳的受试者,其中大多数人还报告注意力不集中或记忆力受损。该试验发现,在180天内每天两次服用2g乙酰L肉碱治疗,不仅提高了整体认知状态筛查(MMSE)的得分,而且还减少了精神和身体疲劳、运动后疲劳、睡眠问题和肌肉疼痛155。一项随机安慰剂对照试验对90名年龄在22岁至66岁之间的健康参与者,检查了一种联合补充剂的效果,该补充剂提供1500mg乙酰L肉碱加15mg长春西汀和150mg石杉碱甲。28天后,补充剂组的整体记忆得分比安慰剂组改善得更多156。
3.2.咖啡因和绿茶:
咖啡因主要来源于咖啡,但也存在于茶叶、瓜拉纳等植物中。咖啡因是提高人类日常生活表现的有效方法。咖啡因是世界上最受欢迎的药物(人类每天总共喝16亿杯),也是进行神经黑客的最受欢迎方法157。咖啡因能提高记忆力、社交能力和警觉性157,158。
咖啡因可以提高认知能力,尤其是在长时间任务中。对大脑黑客感兴趣的人经常将咖啡因作为他们日常养生法的一部分。据认为,咖啡因和茶氨酸可以通过它们的互补作用协同增强大脑功能:咖啡因主要被证明可以提高认知耐力和自我报告的警觉性、觉醒和活力,而L-茶氨酸则被证明可以改善自我报告的放松、紧张和冷静159。一项对六项对照试验数据的荟萃分析发现,单次服用不同剂量的L-茶氨酸加咖啡因可以提高警觉性,并改善某些注意力测试的表现160。例如,一项针对44名18-34岁健康受试者的安慰剂对照试验发现,97mg L-茶氨酸加40mg咖啡因可以在高要求的认知任务中提高注意力,同时提高自我报告的警觉性,减少疲劳161。另一项试验发现,在27名健康的年轻参与者中,50mg咖啡因加100mg L-茶氨酸在90分钟内改善了记忆力、注意力、速度和准确性162。然而,一项包括24名健康年轻人的安慰剂对照交叉试验发现,添加50mg L-茶氨酸似乎可以逆转75mg咖啡因在给药后30分钟对脑血流量和认知能力的短期积极影响163。
3.3.银杏叶:
银杏是一种广泛使用的药用植物,以改善血液流动和降低高血压而闻名。许多临床试验已经检验了它在支持健康大脑功能、减缓认知能力下降和改善痴呆症方面的潜在作用,但结果参差不齐164。对不同类型参与者进行的随机对照试验以及使用不同剂量的不同银杏叶提取物的荟萃分析无法表明银杏叶治疗对记忆力有显著影响165。然而,一项临床研究综述发现,当每天240mg或更多剂量、持续24周或更长时间,以及在患有轻度痴呆的老年人中使用特定的银杏叶提取物时,更有可能增强认知能力164。
据报道,在治疗后的2.5-6小时内,健康年轻受试者单剂量服用120-360mg的银杏叶后,认知功能有益处166,167。尽管许多试验发现,在健康个体中长期使用银杏没有益处168-172,值得注意的是,几项临床试验发现,补充银杏叶2至6周可以改善健康中年女性的认知能力,特别是在复杂或苛刻的记忆任务中173-175。在一项对30名年龄在41至83岁之间的健康参与者进行的试验中,在健康的老年参与者中,每天两次服用120mg银杏叶,持续28天,增加了与复杂视觉信息处理相关的脑电信号,但没有增加年轻参与者176。
几项临床试验评估了单剂量银杏叶和人参提取物组合对健康年轻人的急性影响,并报告了补充后6小时内认知能力的改善177-179。在一项安慰剂对照试验中,256名健康的中年参与者连续12周每天服用320mg的银杏加人参组合后,记忆力得到了改善,这种改善在补充结束后持续了两周180。
3.4.碧萝芷:
来自法国海松树皮的碧萝芷富含多酚,具有广泛研究的清除自由基和抗炎作用,并已被证明具有广泛的抗衰老作用。临床试验表明,它也可以增强健康成年人的认知功能181,182。在一项为期八周的试验中,53名健康大学生每天服用100mg碧萝芷,在注意力、记忆力、执行功能和情绪测试中的表现有所改善。此外,服用碧萝芷的学生的认知测试结果比一组没有服用补充剂的学生的认知测试结果更好183。
在一项对照试验中,59名年龄在35-55岁之间的高氧化应激水平的健康专业人员,那些每天服用150mg碧萝芷12周的人与未治疗的对照组相比,循环自由基水平降低,情绪、注意力和记忆力有了更大的改善184。另一项试验检测了44名年龄在55-70岁、氧化应激水平高的健康参与者在12个月内每天服用100mg碧萝芷的效果,并将他们与没有服用碧萝芷的类似个体进行了比较;碧萝芷治疗组的氧化应激水平降低,多项认知能力改善,而未治疗组则没有185。在一项针对101名年龄在60-85岁之间的健康老年人的试验中,他们每天服用150mg碧萝芷,持续三个月,与服用安慰剂的对照组相比,氧化应激和记忆的测量结果也有所改善186。碧萝芷也被证明对轻度认知障碍患者有益187。
3.5.肌酸:
肌酸是一种存在于全身细胞中的氨基酸衍生物,对肌肉、骨骼和大脑健康具有潜在的积极作用188。肌酸对肌肉力量和功能的益处已得到证实,并因其增强运动能力而被公认189。尽管它主要在肝和肾中合成,但大脑也能够制造肌酸,提示其对脑细胞的重要性。肌酸在细胞能量(三磷酸腺苷或ATP)的产生中起着重要作用,尤其是当能量需求很高时,如运动、复杂的脑力劳动、睡眠不足和一些神经系统疾病189,190。

3.6.磷脂酰胆碱:
这是膳食胆碱的主要来源,存在于蛋黄、奶制品、大豆食品和花生等食物中195。一项对2492名男性进行了近22年跟踪调查的观察性研究发现,磷脂酰胆碱摄入量最高的人患痴呆症的风险比摄入量最低的人低28%。此外,一部分接受认知测试的参与者发现,磷脂酰胆碱摄入量越高,认知能力越好196。
3.7.胞二磷胆碱(CDP胆碱):
它作为胆碱来源的补充剂。临床证据表明,CDP胆碱可能比磷脂酰胆碱更有效地补充胆碱。此外,CDP胆碱中的胆碱可能不太容易转化为三甲胺N-氧化物(TMAO),这是一种由肠道细菌产生的化合物,与动脉粥样硬化有关197。一项安慰剂对照试验,包括100名50-85岁的健康男性和女性,存在与年龄相关的记忆衰退,发现每天服用500mg胞胆碱、持续12周,记忆力测试的表现有所改善198。另一项试验包括40名健康志愿者,连续2周每天服用500mg胞二磷胆碱,比安慰剂更好地降低了氧化应激标志物的水平,改善了反应时间和记忆力的测试得分199。在一项对75名健康男性青少年进行的安慰剂对照试验中,每天补充250mg或500mg胞二磷胆碱28天,可提高注意力和反应速度,减少冲动200。
4.抗压、维持大脑功能:

4.1.茶氨酸:
L-茶氨酸是一种非必需氨基酸,主要存在于绿茶和红茶中,具有增强情绪和认知的作用,并可能成为大脑黑客营养素。随机对照试验的荟萃分析表明,每天补充200-400mg L-茶氨酸可以减轻压力和焦虑症状201。这些影响可能部分归因于其通过影响神经递质信号来调节神经活动的能力202。一些证据表明,在50分钟后的心理任务中,服用L-茶氨酸可以按比例调节脑电活动,且剂量高达400mg203。
一项针对30名健康志愿者的随机对照试验发现,连续四周每天服用200mg L-茶氨酸,可以提高语言流利性(说出特定类别单词的能力)和执行功能测试的得分,而基线得分较低的人的得分更显着。此外,在测量抑郁、焦虑、压力和睡眠症状的量表上,服用L-茶氨酸的人的得分都有所改善204。一项针对50名中老年人的对照试验发现,100mg L-茶氨酸持续12周,可以提高注意力,从而增强工作记忆和执行功能205。
4.2.南非醉茄:
南非醉茄(又称印度人参)是印度阿育吠陀医学的常用草药之一,可以使身体恢复活力,促进所有组织的健康。南非醉茄被认为是一种适应原(Adaptogen),这意味着它可以缓解压力反应并增加适应力(Resilience)206。
在一项针对60名中度至高度感知压力水平的健康成年人的对照试验中,那些每天服用225mg或400mg南非醉茄 30天的人在试验结束时的各种认知测试中表现比试验开始时更好,尽管这些改善与安慰剂没有显著差异207。一项安慰剂对照试验对125名健康参与者进行了研究,这些参与者报告了中度至高度的感知压力水平,结果发现,服用300mg缓释南非醉茄制剂90天,可以改善记忆和注意力,降低皮质醇水平,改善睡眠质量和心理健康208。
4.3.红景天:
红景天是一种主要生长在北极地区的植物,具有公认的适应原作用。红景天历来被用于提高身体耐力、工作效率、寿命和对高原疾病的抵抗力,并用于治疗疲劳、抑郁、贫血、阳痿、胃肠道疾病、感染和神经系统紊乱209。多项临床前研究表明,红景天可能通过增加脑代谢、抑制细胞凋亡和乙酰胆碱分解、减少炎症和氧化应激等作用来改善学习和记忆210。
在一项针对112名健康年轻成年志愿者的安慰剂对照试验中,那些每天服用500mg红景天并持续10天的人在涉及记忆的认知测试中的表现有小到中等的提高;每天服用60mg银杏叶的患者也有类似的改善;每天服用500mg红景天加60mg银杏叶的患者的认知能力比单独服用草药或安慰剂的患者有更大的改善211。在一项对80名认知健康、轻度焦虑的受试者进行的开放标签对照试验中,那些每天两次接受200mg红景天治疗的受试者报告的焦虑、愤怒、压力、困惑和抑郁比没有接受治疗的受试者少。然而,与那些没有接受治疗的人相比,他们的认知能力没有任何改善212。
4.4.复合维矿素:
据报道,多种维生素/矿物质补充剂可改善营养状况不足或亚临床神经或心理症状的成年人的心理健康、感知压力水平、体力和能量水平,以及认知功能的某些方面213。临床研究表明,补充高剂量维生素B族可降低氧化应激和炎症,支持髓鞘形成,并可通过降低同型半胱氨酸水平来改善情绪,同型半胱氨酸是一种与认知障碍有关的氨基酸214-217。
在一项大型随机对照试验中,2262名平均年龄73岁的老年人接受了商业复合维生素/矿物质补充剂、500mg可可黄烷醇或安慰剂治疗,连续三年每天服用。他们还通过电话采访接受了年度认知评估。试验结束时,与安慰剂相比,复合维生素/矿物质组在记忆力、执行功能和整体认知方面都有所改善,而且这种效果在心血管疾病患者中更为明显218。
一项随机试验比较了60名60岁以下健康受试者分别服用复合维生素/矿物质补充剂、维生素D或维生素C持续8周后认知功能的变化,接受复合维生素/矿物补充剂的受试者在工作记忆、学习、运动计划和视觉策略测试中有更大的改善219。在一项针对108名30-70岁健康成年人的随机安慰剂对照试验中,服用提供高剂量B族维生素的复合维生素六个月,增加了大脑关键区域之间的功能连接,这些区域被认为与奖励和情绪处理相关的认知功能有关,尽管没有进行认知测试220。
尽管多种维生素对大脑健康有一些潜在的积极影响,但几项试验没有注意到在健康女性中补充多种维生素/矿物质对认知有任何益处221,222。一项针对97名50岁以下健康女性的试验比较了两剂复合维生素与安慰剂的效果。八周后,那些服用多种维生素的人在认知测试中增加了能量消耗;然而,补充组和安慰剂组的在认知方面表现没有差异223。
4.5.甘磷酰胆碱(α-GCP):

4.6.磷脂:

营养神经黑客

- 提高注意力、思维耐力和精力;和/或
- 减少精神压力和脑雾(思维不清晰等)。
在大脑黑客的背景下,经常讨论的两个重要概念是神经可塑性和认知储备或Resilience(快速恢复能力,或适应力)。
- 神经可塑性:通过加强或形成新的连接,以及修剪或消除未使用的连接来微调神经连接的能力。这一过程有助于有效地学习和适应不同的环境18。
- 认知储备:用于描述大脑通过神经元网络的替代途径传递信息的能力。一个由冗余或可互换的神经元网络组成的强大系统,可以作为一种资源,用来提高大脑功能的速度,或者在通路失效时提供补偿或恢复认知功能的替代通路;认知储备越高,认知能力或认知表现就越好。一般认为,在复杂的环境中度过一生的各种经历可以扩大认知储备,增加在面对压力或神经元损伤等挑战时做出反应和发挥作用的选项数量18,19。
个体的神经黑客目的可能部分取决于年龄。例如,老年人通常关心神经保护和保持健康的大脑功能,而年轻人通常更专注于通过促智剂和其他大脑黑客技术来提高心理表现和耐力。
神经黑客的综合方法依赖于各种互补的策略,包括饮食、生活方式调整,促智药或补充剂的使用,大脑训练活动和游戏,以及旨在提高大脑健康的神经技术(如电刺激装置)2。
神经黑客与天然促智剂
为了更好地理解本文所介绍的研究,首先了解科学家用来评估任何给定干预措施的促智潜力的常用方法是有帮助的。科学家在促智药研究中使用了各种认知评估工具。这些可能包括整体认知测试,如简易精神状态检查(MMSE),尤其是在筛查认知障碍或痴呆症时。更具体的测试通常作为“蓄电池”(Battery)一起进行,提供有关认知功能的详细信息,通常更有助于评估健康受试者的认知表现。认知测试通常用于评估几个特定的认知领域,包括如下20:

本文中的营养素和植物药(提取物)已被证明可以改善健康人大脑或认知功能的一个或多个方面。需要注意的是,由于本文专注于对健康受试者认知功能的研究,一些通常与大脑健康相关的营养素(如甲钴胺、低剂量锂等)可能不会出现在这个列表中。这是因为支持这些营养素的研究通常涉及有潜在神经精神疾病的人,而不是健康人。
值得注意的是,“营养神经黑客”应被视为一种整体方法,强调健康饮食和整体营养,并结合有针对性的营养补充。例如,2022年11月发表的一项随机安慰剂对照试验发现,在习惯性健康饮食的参与者中,补充12周的维生素B族成员、假马齿苋和银杏叶可以改善注意力表现和情绪的某些方面,但在不健康饮食的参与者中则难于产生预期的效果22。
以下营养和植物药(20多种)已经过临床研究,可作为促智剂,基本分为四个类别:

1.1.假马齿苋:
假马齿苋是印度阿育吠陀医学传统药草,可以增强记忆力、减少焦虑和调节血糖水平23。在健康成年参与者中进行的临床试验表明,与安慰剂相比,每天300mg假马齿苋提取物持续90天至12周,可以提高记忆力和情绪测试、视觉处理速度、学习率和一般认知功能的得分24-26。
在一项包括60名医学生的随机安慰剂对照试验中,连续六周每天两次服用150mg假马齿苋标准提取物,可改善注意力和工作记忆测试的表现27。一项包括了60名老年参与者的试验发现,与服用安慰剂的人相比,每天300或600mg的剂量持续12周,在记忆、注意力和认知处理测试中表现更好,表明工作记忆有所改善28。假马齿苋还抑制乙酰胆碱酯酶活性,这种作用会调节神经递质乙酰胆碱的可用性,并与神经保护有关29。此外,在停止补充后四周的随访测试中,假马齿苋对认知的益处持续存在28。一项对28名55岁以上的健康成年人进行的随机安慰剂对照试验发现,与给予安慰剂加认知训练的人相比,那些每天服用320mg假马齿苋提取物、持续12周,每周进行3小时认知训练的人,反应速度较慢,但认知测试的准确性更高30。
一些证据表明,假马齿苋可能具有即时、短期的促智作用,这使得它针对那些对大脑黑客感兴趣的人很有吸引力。在一项有17名健康参与者的小型交叉试验中,与安慰剂相比,在单剂量320mg或640mg标准化高浓度假马齿苋提取物后一小时和两小时进行的多任务认知测试中,表现有所改善。在这项试验中,假马齿苋还降低了唾液皮质醇(体内主要应激激素)的水平,并对情绪产生积极影响,这表明假马齿苋可能在一定程度上起到适应原(增加应激适应性的物质)的作用31。在一项类似的试验中,24名健康志愿者在服用一剂320mg假马齿苋提取物后不久,在重复认知测试中的持续表现优于640mg假马齿苋或安慰剂32。
1.2.蓝莓:
蓝莓以其富含花青素的多酚类物质而闻名。大量研究和临床试验表明,蓝莓提取物和花青素可以增强健康人的认知功能。在一项系统综述中,包括10项在健康成人和儿童中使用蓝莓提取物的对照试验和5项在轻度认知障碍的老年人中使用的对照试验,发现主要的认知效果是记忆改善33。另一项对11项在无认知障碍或轻度认知障碍老年人中进行的试验的综述发现,蓝莓干预可以改善记忆、工作记忆,执行功能、处理速度、注意力以及脑血流量34。一项对六项随机对照试验的荟萃分析发现,蓝莓补充剂显著改善了一项工作记忆测试的表现35。
一些临床证据表明,蓝莓补充剂对认知功能有敏锐的益处。一项针对35名健康中年人的安慰剂对照交叉试验发现,单次服用25g冻干野生蓝莓粉可以改善认知功能测试的表现,尤其是那些涉及更高要求的心理任务的测试36。在一项包括30名健康大学生的安慰剂对照试验中,那些从蓝莓和葡萄中摄入600mg多酚的人在补充后90分钟开始的长时间、高要求的心理测试中表现更好37。
1.3.类胡萝卜素:
叶黄素和玉米黄质是类胡萝卜素家族中的脂溶性营养素。它们高度集中在黄斑,黄斑是眼睛视网膜中负责中央视觉的区域,在那里它们可以保护自身免受氧化应激和短波蓝光的损害38。它们在脑组织中的积累也比其他类胡萝卜素多,因此,叶黄素和玉米黄质占大脑类胡萝卜素含量的66–77%,在减少自由基和减轻神经炎症方面发挥着重要作用39。多项研究表明,在健康的年轻人和老年人中,这些类胡萝卜素的摄入量和浓度越高,患阿尔茨海默病的风险越低,认知功能越好38,40。健康人的脑成像对老年人研究发现,在一年内每天服用10mg叶黄素和2mg玉米黄质,可以增加学习和休息状态(一种神经可塑性的测量方法)下的大脑活动,以及对补充剂反应更好的个体的大脑结构、大脑灰质(主要由神经细胞体组成)体积下降较少41-45。
一项对9项随机对照试验的荟萃分析发现,在健康成年人中,补充叶黄素、叶黄素加玉米黄质、虾青素或β-胡萝卜素可以改善各种认知指标46。例如,一项在59名18-25岁的年轻健康成年人中进行的随机安慰剂对照试验比较了13mg或27mg叶黄素、玉米黄质,以及一种名为消旋玉米黄质(Meso-zeaxanthin)的相关类胡萝卜素和安慰剂。研究人员发现,六个月后,补充类胡萝卜素会降低炎症细胞因子(白细胞介素-1β)的水平,增加脑源性神经营养因子(BDNF)的水平和抗氧化能力,并在记忆、注意力和处理速度测试中表现更好47。类似地,一年后对51名18-30岁的健康受试者进行的安慰剂对照试验,发现可改善记忆力、推理能力和复杂注意力的测试48。在一项随机安慰剂对照试验中,90名40-75岁的健康参与者报告了记忆力、注意力或专注力方面的问题,10mg叶黄素加2mg玉米黄质,每天一次,持续六个月,改善了与学习和记忆相关的某些认知测试的表现39。一项针对91名黄斑类胡萝卜素浓度较低的18岁至60岁健康人的试验发现,一年内补充10mg叶黄素、2mg玉米黄质和10mg消旋玉米黄质比安慰剂更能提高记忆测试的表现49。
1.4.姜黄素:
姜黄素具有抗炎和减少氧化应激的特性,在体内具有广泛的药理作用,并能穿过血脑屏障,使其具有保护神经系统的潜力50。一项2021年的荟萃分析使用了8项随机对照试验的数据,共有389名参与者,其中4项试验针对健康老年参与者,二项试验针对阿尔茨海默病患者,二项研究针对精神分裂症患者。纳入的试验使用了不同剂量的不同类型的姜黄素制剂。分析发现,总的来说,姜黄素治疗至少八周可以改善工作记忆51。另一项荟萃分析表明,姜黄素在改善健康老年人的认知功能方面可能比减轻阿尔茨海默病或精神分裂症患者的症状更有用52。
一项随机安慰剂对照试验在60名健康老年志愿者中进行,他们接受了400mg提供80mg姜黄素的脂质制剂,一小时后发现该制剂改善了注意力和记忆力。此外,连续四周每天服用姜黄素补充剂可以改善工作记忆和情绪测试的表现53。在一项安慰剂对照试验中,80名50至80岁的参与者每天服用相同剂量和形式的姜黄素,持续12周,改善了工作记忆,降低了疲劳54。然而,另一项安慰剂对照试验在39名年龄在45岁至74岁之间的健康男性和女性中进行,发现12周后服用更高剂量(每天2000mg)的相同姜黄素制剂(提供400mg姜黄素)对认知没有益处55。
在40名年龄为51-84岁的受试者中,与安慰剂相比,每天两次服用90mg不同生物可利用形式的姜黄素,持续18个月,可以提高记忆力和注意力测试的表现,还可以减少大脑中异常淀粉样蛋白和tau蛋白(与阿尔茨海默病有关的蛋白质)的积累56。然而,一项包括96名健康老年参与者的试验发现,在12个月内保持认知功能方面,每天三次500mg的另一种姜黄素制剂并不比安慰剂好57。
1.5.野燕麦:
绿燕麦历来被用于治疗焦虑和失眠,临床前证据表明它可以对大脑功能产生积极影响58。在实验室研究中,野燕麦提取物被发现能抑制两种对心理和认知健康有作用的酶:单胺氧化酶-B(MAO-B)和磷酸二酯酶(PDE)59。抑制MAO-B会导致多巴胺等神经递质水平升高,而PDE抑制会导致神经递质信号传递的变化60,61。
在一项针对20名健康参与者的对照试验中,与安慰剂相比,单次800mg剂量的绿燕麦提取物改善了工作记忆、处理速度和注意力测试的表现,并改变了大脑电信号62。一项针对自我报告感觉记忆力下降的健康中年人的试验也显示,在服用800mg(而不是1600mg)绿燕麦提取物后的6小时内,他们在速度、记忆力和执行功能的测试中,认知能力得到了显著改善63。另一项试验包括132名健康个体,他们每天服用430mg、860mg或1290mg绿燕麦提取物或安慰剂,持续29天。补充第一天的认知测试显示,1290mg绿燕麦对工作记忆和多任务处理有急性益处,在第29天,服用430 mg或1290 mg的人有了更大的改善58。然而,在37名参与安慰剂对照交叉试验的健康老年人中,每天1500mg绿燕麦提取物,持续12周,未发现对认知功能测试得分有影响64。
1.6.猴头菇:
猴头菇(又称狮鬃菇)是一种药食同源的菌菇。据报道,它对健康有广泛的益处,包括增强大脑功能,缓解焦虑、抑郁和疲劳65。在一项针对34名50岁以上健康人的随机安慰剂对照试验中,连续12周每天服用4粒800mg的猴头菇粉胶囊,可提高MMSE评分,MMSE是一种衡量认知状态的指标66。其他临床研究表明,猴头菇可能有利于改善轻度认知障碍患者的认知功能67,并有利于保持早期阿尔茨海默病患者的认知能力68。
1.7.中链脂肪酸:
虽然最常见的脂肪由长链脂肪酸组成,但中链甘油三酯(MCTs)由中链脂肪酸构成,如从椰子油或棕榈油中提取的辛酸和癸酸。MCTs可以直接代谢为能量或转化为酮体,为脑细胞提供能量。研究发现,食用强化MCTs的食物可以改善虚弱和认知受损的老年人的认知功能69,70。一项为期四周的对照试验包括30名健康的年轻人,发现与服用安慰剂的人相比,服用每天提供12或18g MCTs饮料的人在2-3周后评估执行功能和注意力的测试中表现更好71。在一项对照试验中,32名健康的老年受试者在饮食中添加MCT,从每天6g开始,经过三周的时间增加到每天18g,而31名类似的受试者则不服用MCT。三个月后,补充MCT的参与者改善了行走平衡,可能表明脑-肌肉交流得到增强72。
MCTs也可能对大脑功能产生直接的积极影响。一项对照交叉试验发现,20名健康的老年志愿者在吃了一顿富含MCT的饭(提供19.9g MCT)后,在注意力的认知测试中表现比吃了富含长链甘油三酯的饭更好73。一项针对19名60岁以上认知健康个体的类似试验发现,与不含MCTs的膳食相比,含20g MCTs膳食后的注意力、执行功能和整体认知功能得分更高74。
1.8. 欧米伽3脂肪酸:

一项临床试验评估了ω-3脂肪酸水平与认知功能之间的关系,该试验包括来自Framingham心脏研究的2183名无痴呆症患者。更高的ω-3指数(定义为红细胞中DHA和EPA的浓度)与海马体积增加和更好的抽象推理有关。EPA和DHA分别维持了这些观察结果。在一项检查APOE-e4基因型非携带者的亚组分析中,表明对阿尔茨海默病易感性增加的遗传标志物、较高的DHA浓度或ω-3指数得分与较大的海马体积有关。相反,在APOE-e4携带者中,较高的EPA浓度与更好的抽象推理有关,较高的ω-3指数与较低的白质高信号负荷有关,这可能与某些脑血管问题的较低风险相对应82。
尽管临床试验的结果好坏参半,但一项对15项随机对照试验数据的荟萃分析发现,补充DHA,无论是否添加EPA,都能改善认知能力,尤其是对那些有轻微记忆问题的人。这些益处需要每天大于1g的EPA/DHA组合剂量和大于580mg的DHA剂量才能显著83。例如,在一项安慰剂对照试验中,176名18至45岁的健康成年人的DHA饮食摄入量较低,在六个月的时间里,每天补充1.16g DHA可以提高反应时间和记忆力的测试成绩84;然而,在一项针对18-35岁健康参与者的试验中,低剂量450mg DHA加90mg EPA或200mg DHA加300mg EPA,持续12周,并不能改善认知能力或情绪参数85。此外,荟萃分析后进行的两项随机安慰剂对照试验发现,尽管剂量和试验持续时间足够,但补充DHA和EPA对健康成年人或有自我报告记忆问题的老年人的认知功能没有影响86,87。
一项对照试验包括285名因冠状动脉疾病而有认知障碍高风险的受试者,研究了高剂量EPA和DHA的可能益处;在补充1860mg EPA和1500mg DHA 12个月后,发现语言流利性、语言、记忆和视觉运动协调性都有所改善,这些改善一直持续到试验结束时的30个月88。
有趣的是,并不是所有的临床试验都表明DHA比EPA更具有益智功效。在一项针对310名50岁以下健康成年人的随机安慰剂对照试验中,那些连续26周每天服用900mg EPA加360mg DHA补充剂的人在记忆速度和准确性方面的所有测试中都比服用900mg DHA加270mg EPA或安慰剂的人有更大的改善89。
1.9.吡咯喹啉醌(PQQ):
PQQ是一种关键的营养素和强效抗氧化剂,有助于多种细胞功能,如线粒体生物发生和基因表达90。临床前研究表明,PQQ具有神经保护特性,并已被发现支持神经元生长,减少活性氧(ROS)的产生,抑制谷氨酸诱导的中风91-93。临床证据表明,PQQ有助于改善健康人的认知功能。
在一项针对41名健康受试者的研究中,每天一次服用20mg PQQ或安慰剂,持续12周,服用PQQ的受试者在注意力和记忆的认知测试中得分显著提高,前额叶皮层的血流量也有所改善,与安慰剂相比94。一项对照试验将64名40-80岁的健康受试者随机分组,每天接受21.5mg PQQ或安慰剂治疗,发现服用PQQ的受试者记忆力、反应时间、注意力和执行功能方面都有显著改善95。另一项研究发现,与安慰剂相比,20-40岁的成年人每天服用20mg PQQ,持续12周,8周后处理速度和执行速度显著提高96。
1.10.丝氨酸磷脂:
可以由磷脂酰胆碱或磷脂酰乙醇胺合成,但更容易由富含DHA的磷脂酰胆碱和磷脂酰乙醇酰胺产生77,97。磷脂酰丝氨酸支持认知功能,如短期和长期记忆、学习、注意力、解决问题、语言和交流,以及神经肌肉活动,如快速运动反应和反射。大脑的磷脂酰丝氨酸含量似乎随着年龄的增长而减少97。在有自我报告记忆问题的老年人中进行的几项临床试验发现,每天补充300mg丝氨酸磷脂可以改善记忆和认知功能的其他方面98-101。此外,在18名健康男大学生中进行的一项安慰剂对照交叉试验发现,在一项名为“连续七次”的认知测试中,每天400mg丝氨酸磷脂持续14天,可以提高速度和准确性。在认知测试中,测试对象被要求从100开始,以7为一组向后数102。
1.11.芳香药草:
迷迭香(Rosmarinus officinalis)、留兰香/绿薄荷(Mentha spicata,Spearmint)和欧薄荷(Mentha piperita,通称薄荷)对大脑功能的影响已被研究。据认为,迷迭香酸(Rosmarinic acid),一种在这些和其他薄荷科植物中发现的芳香酚类化合物,可能通过其穿越血脑屏障并发挥抗炎和清除自由基作用的能力来促进神经保护103。此外,临床前研究表明,迷迭香酸和来自迷迭香、绿薄荷和薄荷的精油与神经递质受体相互作用,可能抑制乙酰胆碱酯酶的活性,乙酰胆碱酯酶是一种分解乙酰胆碱酯酶的酶104,105。
1.11.1.迷迭香:
在一项对80名健康成年人进行的对照试验中,与摄入等量的白开水相比,摄入250ml注入迷迭香的水可以改善认知能力,增加大脑的摄氧量106。一项对28名老年人进行的安慰剂对照交叉试验发现,在评估记忆速度的测试中,单次服用750mg迷迭香叶粉 (这一剂量被认为反映了可行的烹饪摄入量)后的6小时内,记忆速度有所改善,但在服用6000mg迷迭香叶粉后的数小时内,记忆速度恶化107。
还研究了迷迭香的持续使用对认知的影响。在一项对44名健康成年人进行的随机安慰剂对照试验中,发现在63岁以下的一组参与者中,服用含有迷迭香、鼠尾草和香蜂草的酒精提取物两周可以改善短期记忆,但对年龄较大的参与者则没有效果108。在78名健康大学生的随机安慰剂对照试验中,发现每天两次500毫克迷迭香,持续一个月,可以改善记忆力、情绪和睡眠109。然而,在一项试验中,40名低能量的健康年轻受试者在接受认知测试前一小时服用1700mg迷迭香与2000mg黑胡椒或安慰剂,未发现草药组合会影响警觉性或认知能力。有趣的是,参与者戴着鼻夹,以消除迷迭香可能产生的芳香效应110。
芳香疗法是实现迷迭香和其他芳香草药对大脑有益的另一种可能方法。一项针对144名健康志愿者的对照试验报告称,迷迭香精油可以扩散到大气中,改善记忆力和警觉性111,112。在一项包括20名健康志愿者在内的试验中,在散发迷迭香香气的环境中进行认知测试,可以提高速度和准确性113。
1.11.2.留兰香/绿薄荷:
一种富含迷迭香酸的留兰香提取物已成为研究其潜在益智作用的几项临床试验的主题。在一项针对142名年轻健康参与者的安慰剂对照试验中,那些在90天内每天服用900mg标准化为含有14.5%迷迭香酸和24%总酚类化合物的留兰香提取物的人,在测量注意力方面的认知测试中的得分比那些服用安慰剂的人有更大的改善114。在补充相同剂量的高迷迭香酸留兰香提取物后,反应敏捷性,即对刺激快速有效反应的能力也得到了增强,但在安慰剂后没有115。在一项安慰剂对照试验中,60名患有年龄相关记忆障碍的老年参与者,同样的治疗也被发现可以改善一些工作记忆测试以及自我报告的入睡能力116。
1.11.3.薄荷:
在对24名健康年轻参与者进行的安慰剂对照试验中,在单次100ml剂量的薄荷精油胶囊后的3小时内,他们在高要求认知任务中的表现得到改善,长时间认知活动后的精神疲劳有所减少104。在一项对144名健康参与者进行的对照试验中,发现使用薄荷精油进行芳香疗法可以提高记忆力和警觉性111。
2.专注和注意力,缓解脑雾:
可以缓解脑雾(思维不清晰、脑子不畅),增强注意力和任务执行。
2.1.镁:
镁参与许多细胞活动,包括能量生产和蛋白质和DNA的合成。通过参与并促进其他离子穿过细胞膜的运输,镁也有助于调节肌肉细胞收缩和神经细胞兴奋性,保护神经细胞免受可能导致神经细胞死亡的过度兴奋117-119。尽管血镁水平趋于正常,但衰老与体内总镁含量下降有关117。有研究认为,较低的镁浓度会导致慢性炎症和氧化应激,并且是许多与年龄相关的健康问题的一个因素117,120。

特别是L-苏糖酸镁已被证明比其他形式的镁更容易穿过血脑屏障,临床前研究表明,它具有神经保护作用,减少神经炎症,抑制脑组织中的β淀粉样蛋白沉积,增强大脑功能,减缓认知能力下降117,124-126。一项随机安慰剂对照试验,包括44名受试者,年龄在50至70岁之间,有记忆和注意力不集中、焦虑和睡眠问题的患者发现,12周的L-苏氨酸镁补充剂,根据体重每天1500至2000mg,相对于基线和安慰剂,显著改善了整体认知能力127。
2.2.鼠尾草:
鼠尾草(Salvia officinalis和其他品种)是一种富含多酚的植物,历史上一直被用于改善记忆、加快感官、增强大脑功能和延缓认知衰退128。鼠尾草的活性成分已证明具有减少炎症信号、降低氧化应激、调节神经递质水平、减少淀粉样蛋白诱导的神经毒性等作用,以及增加BDNF的释放128-130。在健康年轻人中进行的几项临床试验表明,不同剂量的鼠尾草提取物(S.officinalis或S.lavandulaefolia)可以对记忆和注意力以及情绪产生短期的积极影响128。
在一项针对26名健康运动员的安慰剂对照交叉试验中,在疲劳运动前两小时服用单一600mg剂量的鼠尾草提取物,其中含有S.officinalis和S. lavandulafolia的化合物,也发现可以降低感知耗力的运动水平,提高运动后的记忆测试分数131。一项针对65岁以上成年人的安慰剂对照交叉试验比较了单剂量(167mg、333mg、666mg或1332mg)标准化鼠尾草提取物在给药后1、2.5、4和6小时对认知能力的短期影响。试验发现,在年龄较大人群中,333mg的鼠尾草能最大限度地改善记忆力,并提高注意力测试的分数132。长期使用鼠尾草也可能对认知有好处。在一项对94名健康参与者进行的随机安慰剂对照试验中,与服用安慰剂的参与者相比,服用600mg含有相同二种鼠尾草成分的提取物的参与者在治疗后2小时和4小时进行的一些记忆测试中得分更高;经过29天的日常治疗,记忆力的改善更加明显133。
2.3.芒果叶(提取物):
研究了芒果叶及其活性成分芒果苷(Mangiferin)对脑功能和记忆的影响。芒果苷已被证明可以增强大脑中的电信号,并通过减少炎症和氧化应激、支持线粒体功能、调节神经递质活性和保护BDNF发挥神经保护作用134,135。动物研究结果表明,芒果苷可以保护大脑免受睡眠剥夺、衰老和毒素135。在一项随机、安慰剂对照、交叉试验中,包括70名年龄在18岁至45岁之间的健康参与者,在服用300mg标准芒果叶提取物30分钟、3小时和5小时后,整体认知功能有所改善。与安慰剂相比,当补充芒果叶提取物时,参与者表现出更好的记忆力、注意力和执行复杂任务的能力136。
2.4.积雪草:
积雪草(或称雷公根)是一种亚洲药用植物,通过其减少炎症、氧化应激和线粒体功能障碍的能力,已证明具有神经保护作用,同时增加神经元连接137,138。在一项针对20多岁成年人的试验中,发现一种标准化的积雪草提取物可以提高血液胆碱水平139。
在一项安慰剂对照试验中,包括28名健康的老年参与者,连续两个月每天750mg 积雪草提取物改善了工作记忆,并增加了认知任务期间的脑电活动,但每天250mg和500mg都没有这些效果140。一项包括五项随机对照试验的荟萃分析发现,积雪草可以提高警觉性,减少愤怒,从而改善心理功能;然而,现有的研究不足以得出关于其增强认知能力的结论141。
2.5. 二甲氨基乙醇(DMAE):
DMAE是一种胆碱前体,可优化大脑乙酰胆碱的产生,已证明其具有清除自由基的作用142。一项针对80名情绪障碍受试者的安慰剂对照试验发现,含有DMAE的多种营养补充剂可改善情绪,并在某种程度上改变了脑部电活动,表明注意力增加143。

在健康中青年志愿者的安慰剂对照交叉试验中,发现单次剂量为100、200和400mg的西洋参标准化提取物可在1、3和6小时后改善认知能力145,146。例如,在一项包括61名健康青年和中年参与者的安慰剂对照试验中,单次200mg剂量的标准化西洋参提取物在补充后4小时和6小时的工作记忆和注意力测试中表现增强;此外,连续两周每天服用同样的200mg补充剂可以减少精神疲劳,增强情绪,并进一步提高认知测试成绩;而且人参似乎可以调节肠道微生物组的组成,这可能有助于其认知效果147。
韩国参也一直是健康个体临床试验的主题。据报道,在健康的年轻人中,单次服用200-400mg的高丽人参可以改善认知功能,减轻高要求脑力任务时的精神疲劳148-150。一项针对51名健康参与者的试验发现,与安慰剂相比,连续八周每天服用1000mg高丽人参不仅能提高整体认知功能得分,而且还能增加灰质(主要由神经细胞体和其他细胞组成的脑组织)的体积151。在一项交叉试验中,包括50名具有高度职业压力的健康个体,在1天、5天和12天后,每天服用418mg人参皂苷富集制剂的高丽人参比每天服用768mg未富集的高丽人参粉或安慰剂更有效152。
3.缓解脑疲劳、增强认知耐力:
可以强化认知耐力,避免普遍的精神疲劳。
3.1.乙酰L肉碱:
L-肉碱是细胞脂肪代谢和线粒体能量生产所必需的氨基酸。它在体内制造,储存在骨骼肌、心肌和大脑等代谢需求高的组织中。乙酰L肉碱是一种穿过血脑屏障的肉碱,因此可以通过增强神经生长因子、神经递质和神经激素的产生来调节神经活动153。此外,乙酰L肉碱可以帮助恢复神经细胞膜的完整性和功能,增强线粒体功能,并防止神经毒素154。
一项随机安慰剂对照试验包括96名70岁以上且患有疲劳的受试者,其中大多数人还报告注意力不集中或记忆力受损。该试验发现,在180天内每天两次服用2g乙酰L肉碱治疗,不仅提高了整体认知状态筛查(MMSE)的得分,而且还减少了精神和身体疲劳、运动后疲劳、睡眠问题和肌肉疼痛155。一项随机安慰剂对照试验对90名年龄在22岁至66岁之间的健康参与者,检查了一种联合补充剂的效果,该补充剂提供1500mg乙酰L肉碱加15mg长春西汀和150mg石杉碱甲。28天后,补充剂组的整体记忆得分比安慰剂组改善得更多156。
3.2.咖啡因和绿茶:
咖啡因主要来源于咖啡,但也存在于茶叶、瓜拉纳等植物中。咖啡因是提高人类日常生活表现的有效方法。咖啡因是世界上最受欢迎的药物(人类每天总共喝16亿杯),也是进行神经黑客的最受欢迎方法157。咖啡因能提高记忆力、社交能力和警觉性157,158。
咖啡因可以提高认知能力,尤其是在长时间任务中。对大脑黑客感兴趣的人经常将咖啡因作为他们日常养生法的一部分。据认为,咖啡因和茶氨酸可以通过它们的互补作用协同增强大脑功能:咖啡因主要被证明可以提高认知耐力和自我报告的警觉性、觉醒和活力,而L-茶氨酸则被证明可以改善自我报告的放松、紧张和冷静159。一项对六项对照试验数据的荟萃分析发现,单次服用不同剂量的L-茶氨酸加咖啡因可以提高警觉性,并改善某些注意力测试的表现160。例如,一项针对44名18-34岁健康受试者的安慰剂对照试验发现,97mg L-茶氨酸加40mg咖啡因可以在高要求的认知任务中提高注意力,同时提高自我报告的警觉性,减少疲劳161。另一项试验发现,在27名健康的年轻参与者中,50mg咖啡因加100mg L-茶氨酸在90分钟内改善了记忆力、注意力、速度和准确性162。然而,一项包括24名健康年轻人的安慰剂对照交叉试验发现,添加50mg L-茶氨酸似乎可以逆转75mg咖啡因在给药后30分钟对脑血流量和认知能力的短期积极影响163。
3.3.银杏叶:
银杏是一种广泛使用的药用植物,以改善血液流动和降低高血压而闻名。许多临床试验已经检验了它在支持健康大脑功能、减缓认知能力下降和改善痴呆症方面的潜在作用,但结果参差不齐164。对不同类型参与者进行的随机对照试验以及使用不同剂量的不同银杏叶提取物的荟萃分析无法表明银杏叶治疗对记忆力有显著影响165。然而,一项临床研究综述发现,当每天240mg或更多剂量、持续24周或更长时间,以及在患有轻度痴呆的老年人中使用特定的银杏叶提取物时,更有可能增强认知能力164。
据报道,在治疗后的2.5-6小时内,健康年轻受试者单剂量服用120-360mg的银杏叶后,认知功能有益处166,167。尽管许多试验发现,在健康个体中长期使用银杏没有益处168-172,值得注意的是,几项临床试验发现,补充银杏叶2至6周可以改善健康中年女性的认知能力,特别是在复杂或苛刻的记忆任务中173-175。在一项对30名年龄在41至83岁之间的健康参与者进行的试验中,在健康的老年参与者中,每天两次服用120mg银杏叶,持续28天,增加了与复杂视觉信息处理相关的脑电信号,但没有增加年轻参与者176。
几项临床试验评估了单剂量银杏叶和人参提取物组合对健康年轻人的急性影响,并报告了补充后6小时内认知能力的改善177-179。在一项安慰剂对照试验中,256名健康的中年参与者连续12周每天服用320mg的银杏加人参组合后,记忆力得到了改善,这种改善在补充结束后持续了两周180。
3.4.碧萝芷:
来自法国海松树皮的碧萝芷富含多酚,具有广泛研究的清除自由基和抗炎作用,并已被证明具有广泛的抗衰老作用。临床试验表明,它也可以增强健康成年人的认知功能181,182。在一项为期八周的试验中,53名健康大学生每天服用100mg碧萝芷,在注意力、记忆力、执行功能和情绪测试中的表现有所改善。此外,服用碧萝芷的学生的认知测试结果比一组没有服用补充剂的学生的认知测试结果更好183。
在一项对照试验中,59名年龄在35-55岁之间的高氧化应激水平的健康专业人员,那些每天服用150mg碧萝芷12周的人与未治疗的对照组相比,循环自由基水平降低,情绪、注意力和记忆力有了更大的改善184。另一项试验检测了44名年龄在55-70岁、氧化应激水平高的健康参与者在12个月内每天服用100mg碧萝芷的效果,并将他们与没有服用碧萝芷的类似个体进行了比较;碧萝芷治疗组的氧化应激水平降低,多项认知能力改善,而未治疗组则没有185。在一项针对101名年龄在60-85岁之间的健康老年人的试验中,他们每天服用150mg碧萝芷,持续三个月,与服用安慰剂的对照组相比,氧化应激和记忆的测量结果也有所改善186。碧萝芷也被证明对轻度认知障碍患者有益187。
3.5.肌酸:
肌酸是一种存在于全身细胞中的氨基酸衍生物,对肌肉、骨骼和大脑健康具有潜在的积极作用188。肌酸对肌肉力量和功能的益处已得到证实,并因其增强运动能力而被公认189。尽管它主要在肝和肾中合成,但大脑也能够制造肌酸,提示其对脑细胞的重要性。肌酸在细胞能量(三磷酸腺苷或ATP)的产生中起着重要作用,尤其是当能量需求很高时,如运动、复杂的脑力劳动、睡眠不足和一些神经系统疾病189,190。

3.6.磷脂酰胆碱:
这是膳食胆碱的主要来源,存在于蛋黄、奶制品、大豆食品和花生等食物中195。一项对2492名男性进行了近22年跟踪调查的观察性研究发现,磷脂酰胆碱摄入量最高的人患痴呆症的风险比摄入量最低的人低28%。此外,一部分接受认知测试的参与者发现,磷脂酰胆碱摄入量越高,认知能力越好196。
3.7.胞二磷胆碱(CDP胆碱):
它作为胆碱来源的补充剂。临床证据表明,CDP胆碱可能比磷脂酰胆碱更有效地补充胆碱。此外,CDP胆碱中的胆碱可能不太容易转化为三甲胺N-氧化物(TMAO),这是一种由肠道细菌产生的化合物,与动脉粥样硬化有关197。一项安慰剂对照试验,包括100名50-85岁的健康男性和女性,存在与年龄相关的记忆衰退,发现每天服用500mg胞胆碱、持续12周,记忆力测试的表现有所改善198。另一项试验包括40名健康志愿者,连续2周每天服用500mg胞二磷胆碱,比安慰剂更好地降低了氧化应激标志物的水平,改善了反应时间和记忆力的测试得分199。在一项对75名健康男性青少年进行的安慰剂对照试验中,每天补充250mg或500mg胞二磷胆碱28天,可提高注意力和反应速度,减少冲动200。
4.抗压、维持大脑功能:

4.1.茶氨酸:
L-茶氨酸是一种非必需氨基酸,主要存在于绿茶和红茶中,具有增强情绪和认知的作用,并可能成为大脑黑客营养素。随机对照试验的荟萃分析表明,每天补充200-400mg L-茶氨酸可以减轻压力和焦虑症状201。这些影响可能部分归因于其通过影响神经递质信号来调节神经活动的能力202。一些证据表明,在50分钟后的心理任务中,服用L-茶氨酸可以按比例调节脑电活动,且剂量高达400mg203。
一项针对30名健康志愿者的随机对照试验发现,连续四周每天服用200mg L-茶氨酸,可以提高语言流利性(说出特定类别单词的能力)和执行功能测试的得分,而基线得分较低的人的得分更显着。此外,在测量抑郁、焦虑、压力和睡眠症状的量表上,服用L-茶氨酸的人的得分都有所改善204。一项针对50名中老年人的对照试验发现,100mg L-茶氨酸持续12周,可以提高注意力,从而增强工作记忆和执行功能205。
4.2.南非醉茄:
南非醉茄(又称印度人参)是印度阿育吠陀医学的常用草药之一,可以使身体恢复活力,促进所有组织的健康。南非醉茄被认为是一种适应原(Adaptogen),这意味着它可以缓解压力反应并增加适应力(Resilience)206。
在一项针对60名中度至高度感知压力水平的健康成年人的对照试验中,那些每天服用225mg或400mg南非醉茄 30天的人在试验结束时的各种认知测试中表现比试验开始时更好,尽管这些改善与安慰剂没有显著差异207。一项安慰剂对照试验对125名健康参与者进行了研究,这些参与者报告了中度至高度的感知压力水平,结果发现,服用300mg缓释南非醉茄制剂90天,可以改善记忆和注意力,降低皮质醇水平,改善睡眠质量和心理健康208。
4.3.红景天:
红景天是一种主要生长在北极地区的植物,具有公认的适应原作用。红景天历来被用于提高身体耐力、工作效率、寿命和对高原疾病的抵抗力,并用于治疗疲劳、抑郁、贫血、阳痿、胃肠道疾病、感染和神经系统紊乱209。多项临床前研究表明,红景天可能通过增加脑代谢、抑制细胞凋亡和乙酰胆碱分解、减少炎症和氧化应激等作用来改善学习和记忆210。
在一项针对112名健康年轻成年志愿者的安慰剂对照试验中,那些每天服用500mg红景天并持续10天的人在涉及记忆的认知测试中的表现有小到中等的提高;每天服用60mg银杏叶的患者也有类似的改善;每天服用500mg红景天加60mg银杏叶的患者的认知能力比单独服用草药或安慰剂的患者有更大的改善211。在一项对80名认知健康、轻度焦虑的受试者进行的开放标签对照试验中,那些每天两次接受200mg红景天治疗的受试者报告的焦虑、愤怒、压力、困惑和抑郁比没有接受治疗的受试者少。然而,与那些没有接受治疗的人相比,他们的认知能力没有任何改善212。
4.4.复合维矿素:
据报道,多种维生素/矿物质补充剂可改善营养状况不足或亚临床神经或心理症状的成年人的心理健康、感知压力水平、体力和能量水平,以及认知功能的某些方面213。临床研究表明,补充高剂量维生素B族可降低氧化应激和炎症,支持髓鞘形成,并可通过降低同型半胱氨酸水平来改善情绪,同型半胱氨酸是一种与认知障碍有关的氨基酸214-217。
在一项大型随机对照试验中,2262名平均年龄73岁的老年人接受了商业复合维生素/矿物质补充剂、500mg可可黄烷醇或安慰剂治疗,连续三年每天服用。他们还通过电话采访接受了年度认知评估。试验结束时,与安慰剂相比,复合维生素/矿物质组在记忆力、执行功能和整体认知方面都有所改善,而且这种效果在心血管疾病患者中更为明显218。
一项随机试验比较了60名60岁以下健康受试者分别服用复合维生素/矿物质补充剂、维生素D或维生素C持续8周后认知功能的变化,接受复合维生素/矿物补充剂的受试者在工作记忆、学习、运动计划和视觉策略测试中有更大的改善219。在一项针对108名30-70岁健康成年人的随机安慰剂对照试验中,服用提供高剂量B族维生素的复合维生素六个月,增加了大脑关键区域之间的功能连接,这些区域被认为与奖励和情绪处理相关的认知功能有关,尽管没有进行认知测试220。
尽管多种维生素对大脑健康有一些潜在的积极影响,但几项试验没有注意到在健康女性中补充多种维生素/矿物质对认知有任何益处221,222。一项针对97名50岁以下健康女性的试验比较了两剂复合维生素与安慰剂的效果。八周后,那些服用多种维生素的人在认知测试中增加了能量消耗;然而,补充组和安慰剂组的在认知方面表现没有差异223。
4.5.甘磷酰胆碱(α-GCP):

4.6.磷脂:
